I took a few days off of work to hunt a long weekend - 11-8 through 11-13. Thursday morning (11-8) just after dawn, I snuck into a thick spot that I prepped last spring in anticipation of the rut. This was my first time hunting the area this year.
There were a few scrapes with licking branches near the base of my tree, so when nature called I added some homemade scent. Here is a small 6-point yearling that worked a couple of scrapes underneath me and continued on his way (phone pics):
I returned to that tree on the evening of 11-9 when I again had a favorable wind. It was another successful evening as I had a few encounters with rutting bucks and had a shot at a nice two year-old (unfortunate shoulder blade hit = no deer).
I returned to that same tree the next morning for the gun season opener 11-10. Here is a small spike that worked the same scrapes that morning after I had "freshened" the scrape, notice the "oh sh$t" moment when he spots me while working the licking branch:

Here is a trail cam video of a nice 8-point two year-old that worked the same scrapes the next evening (11-11):

While I am not sure that human urine will prove effective on mature bucks, it certainly did not seem to negatively affect these scrapes.
Also, note that I hunted the same tree:
11-8 morning
11-9 evening
11-10 morning
11-13 morning (best morning, multiple nice bucks seen)
I have an extremely effective access route using the river and hip boots for this spot only covering 40 ft of dry ground before I am up the tree. I feel that I can over-hunt this spot with the correct winds during the rut.

I hunt out of a Tree Saddle for over 90% of my hunts. I rarely get busted while hunting in my saddle, not my experience with hang-ons.
I mainly scout, install screw-in steps, and prep all of my trees in the winter / spring. I pack in a LW Assassin for longer hunts as it increases the comfort of the system. I typically do not return to the area until I hunt the tree.
